He was married to Jane Bell Murphy in 1865, and had children Francis Thomas Murphy, Mary Alice Murphy Bales, William Amos Murphy jr, Oswald Robert Murphy, Edith Kate (Murphy) Pettitt, Laura Jane Murphy, and Francis Edward Sylvester Murphy. Amos Murphy died at age 60 years old on August 10, 1905 in Maldon, Mount Alexander Shire County.
