James Deflora was born in New York to parents John A Deflora (1876-1952) and Theresa A Catanzaro (1884-1947). He had multiple full and half siblings including Grace (born 1903), Ralph (born 1906), Florence (1909-1940), Anthony (born 1917), Rosaline Marie (1923-1991), and others. He married Grace Deflora and had sons Noel and Joel. He was a World War II veteran and earned a Bronze Star and Certificate of Merit Citation.

He was a white Italian. Both of his parents were born and raised in Italy. His father, John A Deflora, was born in Naples, Napoli, Campania, Italy.

James was Catholic and his funeral service was held at St. Malachy's Catholic Church in Fort Lauderdale, Florida.

James Deflora enlisted on July 9, 1942 in Fort Jay Governors Island New York (Military serial#: 32402295). His terms of enlistment were, "Enlistment For The Duration Of The War Or Other Emergency, Plus Six Months, Subject To The Discretion Of The President Or Otherwise According To Law". He was discharged on November 17, 1945 and was a WWII veteran.

In 1940, he was employed by "Rockwood and Co" in Brooklyn and later in life James Deflora worked as a railroad conductor under the United Transportation Union.
